屈臣氏水;甲醇为色谱纯;磷酸二氢钾色谱纯;磷酸、四丁基硫酸氢铵为分析纯;
胞嘧啶核苷酸(胞苷酸)、腺嘌呤核苷酸(腺苷酸)、尿嘧啶核苷酸(尿苷酸)、次黄嘌呤核苷酸、鸟嘌呤核苷酸(鸟苷酸)标准品;
高效液相色谱柱:Venusil ® C18 Plus;5μm,120Å,4.6×250mm
样品制备
5种核苷酸混合标准溶液:分别称取核苷酸标准品CMP、AMP、UMP各10mg、GMP、IMP各5mg(精确至0.1mg),用水溶解成并定容至100mL,混匀,即得;
某品牌婴幼儿奶粉样品溶液:称取5.0g奶粉样品,用20mL热水(50~60℃)充分溶解试样,冷却至室温;用10%乙酸溶液调节pH至4.1,移至50mL容量瓶中,定容,滤纸过滤,所得滤液用0.45μm滤膜过滤,即得样品溶液。
色谱条件
色谱柱:
Venusil ® C18 Plus;5μm,120Å,4.6×250mm;
流动相:
称取1.360g磷酸二氢钾,加0.4753g四丁基硫酸氢铵至1000mL容量瓶中,用水补齐至刻度,准确加2.5ml磷酸和40mL甲醇,混匀后超声10min。(备注:由于流动相中含离子对试剂,色谱柱需要先用流动相平衡至少90min后,再运行4针标准品,系统才能稳定)
波长:254nm;
柱温:30℃;
流速:1.0mL/min;
进样量:10µL。
结果与讨论
本实验按照GB5413.40-2016婴幼儿食品和乳品中核苷酸的测定中方法,使用Venusil® C18 Plus色谱柱对5种核苷酸混合标准溶液、婴幼儿奶粉样品溶液进行检测,由以下结果可知,婴幼儿奶粉样品中五种核苷酸与杂质的分离度大于1.5,回收率97%~109%,满足实验需求。
